I've seen some pretty drastic differences on the dyno between different headers. Could be as much as 5% or more, which is 40hp on an 800hp engine or almost 25hp on a 600hp engine.
Here's a graph of a 900hp pump gas big block with a big set of old school Pro Stock Hookers with 2-1/2" x 4-1/4" pipes vs a set of custom 2-1/4 x 3-1/2 Lemons. Biggest difference was 40hp but almost 35 lbs ft and 37+ hp average from 5000-7000 so yeah, headers can make a BIG difference.
