How do you know if you have one and what would be the effects if you used the wrong pump on the wrong block either way?

Just curious.

You can't really mix them up. The water pump on a reverse flow engine is gear driven off the cam. The LT1/LT4 is reverse cooled, and it doesn't have a traditional provision for a distributor either. It's "optispark" is mounted in front of the timing cover, and is crank driven. As far as I know, only the LT1, and LT4 engines were reverse cooled. these are found in 4th gen F bodys from 93-97, and corvettes from 92-96. They were reverse cooled to prevent detonation caused by increased compression.

Do you mean reverse cooled or reverse rotation? Serpentine belt drives turn the W/P in the opposite direction as a V-belt. If you used a pump for one on the other type of drive, you'd get no coolant flow whatsoever.

Reversed cooled. Cool water is introduced to heads first then the block. This allows the compression to be at least a full point higher that a conventional system.
