I'm doing a full body-off restoration on a 68 conv. I'm using a different hood,doors,fenders. Should I re-assemble on the frame for a fit check and trimming before painting? Then dis-assemble again the body and all attaching parts for individual painting of each part in order to get all hidden areas and not have overspray on the frame and supporting parts?

That's EXACTLY what my body-man is doing this week. You don't want to fit the fenders, hood, and doors with fresh paint...

Test fit everything. Including bumpers, grill and trim. be glad you do it now and not when the new paint is on.
I restored my 37 chevy coupe this way. It nearly broke my heart to dissasemble it after everything finally fit right.
A suggestion: before you dissasemble to paint drill small pilot holes in inconspicous places through the sheetmetal and hinges/shims for the various door/lids/fenders etc. Then on reassembly use that size drill by hand to help line up the panels again.

Just a quick note. Found this out the hard way. If you are re-painting with base-coat/clear coat and the paint has metallic in it, beware of painting the individual pieces. I had this done on my 71LS5 and the guy painting it had a heck of a time keeping the "zebra tails" out of the finish (it is Nevada Silver...lots o' metallic). Anyway, the easiest way to take care of that is to assemble doors, fenders, deck, hood, and paint everything together. Otherwise, fog coat may be in order and there is still the possibility of it not looking the way you want it to. You will always wonder why that fender looks darker/lighter than the doors.

I am going through the exact same thing with my car too. I am in the process of fitting all of the sheetmetal back on the car and will then be removing it all again. My bodyman is going to paint everything off the car (a solid color) except for the fenders. After the initial fit check & removal, I am going to spray the "hard to reach" spots on the fenders & reinstall them. The hinges will also remain on the body, in their proper places, for painting.
