Can anyone tell me which hose goes to the intake and which goes to the water pump for a big block? And also does it really matter which goes where?

I usually put the lower hose to the water pump, and the upper hose to the intake.
Then there is also a very short hose that connects the water pump and intake (only about 2" long). This hose is optional though, I didn't hook it up on my car and it works fine.

The hoses are two different diameters. The supply hose from the manifold is 5/8 inch and goes to the smaller fitting on the heater core , while the larger (3/4inch) hose goes on the pump.

Thanks JW. That's exactly what I was looking for. I can't find the 3/4 hose fitting in chrome. All they seem to make is a tall one. Any ideas where to find a short chrome 3/4"?

The bypass hose isn't "optional" although you can run without it in most cases. GM put the hose there for a reason on a big block. It allows coolant to circulate via the water pump until the thermostat opens up. Without some flow through the engine, the block will develop hot spots then be hit with a rush of cooler water when the thermostat opens. (: Many people that block off the bypass hose drill one or two 1/8" holes in the thermostat to allow coolant to circulate.
I'd suggest hooking the hose back up or drilling a couple of holes.
