Just bought a new Edelbrock RPM air gap manifold for the big block. There is a 1/4" opening of the exhaust crossover opening (long slit of an opening above and below the main exhaust crossover port)) that the intake manifold does not cover. Will I need a metal gasket for this occasion or will a regular gasket be enough to seal up this hole? Heads are 781's if that makes a difference.

I don't have a set of 781's to look at, but are you sure the slit is part of the exhaust crossover, and not just a cooling divider ? A lot of Chrysler and Pontiac heads are made like that and that area remains open to the air for cooling. Look and see if it actually goes into the exhaust crossover, or if it is just a recessed area in the head casting with no opening to anything.
