I have a 5/16 line originally from my old inline 6. Well my 350 idles and reves great under no load but onces in gear seems to bog down and not have much throttle response and slow to get going. Checking some threads a lot of you guys have the 3/8 line. Should a 3/8 line over a 5/16 make that much of a difference?

Put a fuel pressure guage and the guessing's over.

No check, timing, blocked filters, carb problems etc. something else is wrong until you get up to higher rpm and more mph then 5/16 will slow you down

fuel pressure is fine new mechanical pump installed

sounds like a carburator problem...

If the fuel pressure's constant at the carb ,it's not the line that's holding you back.

5/16's line was good with stock pump to mid/low 12 in my car.

But did you check the pressure? You have to eliminate the variables. Just because you put a new pump on it doesn't mean it's not the problem.
